In the Lab
The Xie lab is interested in engineering nanoparticles, either nanocrystal-, polymer- or protein-based, for imaging and drug delivery, with a focus on cancer. These involve developing novel materials with unique physical and chemical properties. Moreover, the lab device surface coatings of nanoparticles to impart functionalities and tailor the particles’ roles as imaging/therapy agents.
For instance, the Xie lab have developed a mesoporous silica (MSN)-template calcination technology to synthesize 3 nm gadolinium intercalated carbon dot (Gd@Cdot), compared to Gd complexes, Gd@Cdots afford higher T1 contrast ability and far greater chemical stability. These nanoparticles can be efficiently excreted through renal clearance, minimizing toxicities commonly seen with other heavy metal nanoparticles. Another example is ferritin-based drug delivery. Our lab has modified the protein, both chemically and/or genetically, to present targeting ligands onto the surface. The resulting nanocages can encapsulate a wide range of therapeutics, and deliver them to tumors in a site-specific fashion. The Xie lab is also working on nanoparticle-based photodynamic therapy, radiation therapy, and immune therapy against cancer.
